Regarding these compounds with large 4-position substituents, it is interesting to note that while the 2C-x and DOx versions are antagonists, this may not be the case when you add an NBOMe - if the patents are to be believed, both NBOMe-2C-Am and NBOMe-thiobuscaline are agonists at 5-HT2A, even though the parent compounds are antagonists. Not unprecedented, after all RH-34 is basically an NBOMe version of ketanserin (though it is not psychedelic in humans due to dose-limiting off target side effects), but it does make me curious how broadly this applies. There is an old compound of Glennon's which was iirc 4-(3-phenylpropyl)-3,5-dimethoxyamphetamine which he claimed to surprisingly show partial agonist character despite the bulky 4-position substituent, though it acts like an antagonist in practice as the efficacy is so low. But I wonder if the 2C, NBOMe version of that would be a proper agonist. Meodipt (talk) 22:17, 5 February 2025 (UTC)
